They accused Israel of genocide and urged the court to compel Israel to halt its military operations in Gaza.
Israel is set to present its defence, contending that its actions are a response to Hamas's lethal attacks on October 7.
Prime Minister Benjamin Netanyahu emphasised Israel's commitment to not permanently displacing Gaza's residents or occupying the territory.
Zane Dangor, director general of South Africa's department of international relations and cooperation, acknowledged the gravity of the genocide allegation, stating it is a "strong" but not unfounded claim.
The International Court of Justice may promptly rule on South Africa's plea for Israel to suspend its military campaign, while a final verdict on the genocide accusation could extend over several years.
